Description
The protein encoded by this gene is a putative protein kinase that is induced by the transcription factor NF-kappaB. The encoded protein is a negative regulator of NF-kappaB and can also sensitize cells to TNF- and TRAIL-induced apoptosis. In addition, this protein can negatively regulate the cell survival serine-threonine kinase AKT1. Differential promoter usage and alternate splicing result in multiple transcript variants.
TRIB3 is a member of the Tribbles homolog gene family (TRIB1, \update), all of them are considered to be pseudokinases. Most of its effects TRIB3 mediates by binding to and inhibition of AKT1 signaling [Prudente \emph, Endocr Rev, 2012, 33:526-546]. Other related pathways involved in TRIB3 action include e.g. \update and MAPK8 mediated signaling and \update induced ER-dependent gene expression [Prudente et al., Endocr Rev, 2012, 33:526-546]. TRIB3 has a higher expression in EBOV (MARV)-infected human cell samples 23 h p.i., with a fold change of 1.19 (1.0) compared to Mock 23 h and read\_max values of 682 and 417, respectively. In bat cells, this gene shows an overall lower expression (read\_max: 108 and 42, respectively), although it is also slightly up-regulated 23 h p.i..
